Potential of early diagnosis of coronary artery disease using differential expression of a circulating molecule: a cross-sectional study

Abstract

Background and Aims: Coronary artery disease (CAD) remains the leading cause of mortality and disability worldwide. The Antisense Non-Coding RNA in the INK4 Locus (ANRIL) plays a significant role in the development of atherosclerosis. The aim of this study is to evaluate the potential of this molecule as an early diagnostic biomarker for coronary artery disease by comparing its expression levels between patients with the disease and healthy individuals.
Materials and Methods: In this cross-sectional study, 59 patients with CAD and 26 healthy individuals, were enrolled. Group allocation was based on the angiography reports. Blood samples were subjected to RNA extraction and cDNA synthesis, followed by quantification of the expression levels of both linear and circular forms of ANRIL using real-time PCR.
Results: The findings showed that the expression levels of both the circular and linear forms of ANRIL did not differ significantly between patients with CAD and healthy individuals. However, when the ratio of circular to linear ANRIL expression was calculated, a statistically significant difference was observed between the two groups (P value < 0.05).
Conclusion: Given the significant difference in the ratio of circular to linear ANRIL expression, this parameter may serve as a potential tool for the early detection of CAD through a simple, non-invasive blood test. This approach could substantially contribute to the prevention of morbidity and mortality, while also reducing the overall healthcare burden associated with this prevalent condition.
